Dr. Shiyu Liu earned his Bachelor of Science degree from the School of Life Sciences at Peking University in 2016. He completed his Ph.D. in Computational Biology and Bioinformatics at Duke University in 2021, under the mentorship of Dr. Jason Locasale. Following graduation, Dr. Liu continued as a postdoctoral associate in Dr. Locasale’s lab. His research focuses on developing flux analysis algorithms and software, and applying these tools to advance metabolic research in areas such as nutrition, diet, aging, and cancer biology.
讲座摘要/Abstract:
Metabolism involves a complex network of chemical reactions within living organisms, and isotope labeling is a powerful method for analyzing reaction rates under physiologically relevant conditions. This technique supplies organisms or cells with isotopically labeled substrates, and metabolic activity is captured through labeling patterns, which are quantified by mass spectrometry (MS) or nuclear magnetic resonance (NMR). These patterns are analyzed using flux analysis to determine the metabolic network's fluxes that best fit the data.
However, current flux analysis methods are inefficient, have limited applicability, and lack precision and robustness evaluations, making them less trustworthy and often viewed as complementary in metabolism studies. My contributions to this field focus on three key areas: First, I demonstrated the effectiveness of flux analysis in providing critical insights into metabolism, such as identifying key differential reactions and pathways under various conditions. Second, I expanded its applicability from cultured cells to more complex systems, including isotope-infused mice and isotope-fed fruit flies. Third, I developed a benchmarking system to systematically evaluate precision and robustness, and designed a new pipeline to address the inherent uncertainties of traditional flux analysis methods. Together, these advancements establish flux analysis as a more robust, efficient, and broadly applicable methodology, offering strong support for researchers across diverse biological fields.
